Le β-D-Fructose 1,6-diphosphate (F6DP) est un dérivé du fructose produit au cours de la glycolyse par phosphorylation du fructose 6-phosphate par la phosphofructokinase avec consommation d'une molécule d'ATP. Il est aussi un intermédiaire de la néoglucogenèse où il est produit par l'action de la fructose-disphosphate aldolase, ainsi que dans les voies de biosynthèse du fructose et du mannose.
Régulation du flux glycolytique
Le F6DP est l'un des régulateurs allostérique de l'activité de la phosphofructokinase, l'enzyme qui catalyse sa production au cours de la glycolyse (inhibition par le produit de réaction). Il agit comme un régulateur cybernétique par feedback négatif : L'accumulation de F6DP signifie qu'il n'est pas consommé par les étapes en aval de la voie, et que donc le besoin en énergie est moindre, qu'il faut donc en quelque sorte ralentir la glycolyse.
